Summary

Interferon regulatory factor-1 (IRF-1) can induce retinoic acid-induced gene G (RIG-G) expression. STAT1 enhances this RIG-G induction via the JAK-STAT pathway, but it is not essential for RIG-G expression.

Background:

The IRF-9/STAT2 complex drives RIG-G expression independently of STAT1 and the JAK-STAT pathway.
IRF-1 has been observed to regulate RIG-G induction and IRF-9/STAT2 expression in certain contexts.
The precise mechanisms of IRF-1's action on RIG-G expression remained unclear.

Purpose of the Study:

To elucidate the mechanisms by which IRF-1 influences RIG-G gene expression.
To investigate the role of STAT1 in IRF-1-mediated RIG-G induction.
To understand the interplay between IRF-1, STAT1, and the IRF-9/STAT2 complex in regulating RIG-G.

Main Methods:

Utilized STAT1-deficient U3A cells and STAT1-reconstituted U3A cells.
Analyzed RIG-G gene promoter activity, specifically focusing on IFN-stimulated response elements.
Investigated the expression levels of IRF-9 and STAT2 under different experimental conditions.

Main Results:

IRF-1 induces RIG-G expression in STAT1-deficient cells via IFN-stimulated response elements.
IRF-1 fails to upregulate IRF-9 and STAT2 in STAT1-deficient cells unless STAT1 is reintroduced.
STAT1 significantly enhances IRF-1 or IRF-9/STAT2 complex-mediated RIG-G induction through the JAK-STAT pathway, although STAT1 is not essential for basal RIG-G expression.
In STAT1-expressing cells, IRF-1 indirectly activates RIG-G expression in an IRF-9/STAT2-dependent manner.

Conclusions:

RIG-G expression is independent of the classical JAK-STAT pathway but can be significantly enhanced by it.
IRF-1 can regulate RIG-G expression through both STAT1-dependent and STAT1-independent mechanisms.
This study provides a deeper understanding of the complex regulatory network governing RIG-G gene expression.
