DNA walker-assisted aptasensor for highly sensitive determination of Ochratoxin A

Summary

A novel aptasensor detects Ochratoxin A (OTA) using gold nanoparticles-molybdenum oxide and a DNA walker. This highly sensitive electrochemical biosensor accurately measures OTA in various food and serum samples.

Ochratoxin A (OTA) is a harmful fungal toxin affecting human and animal health.
Accurate detection of OTA is crucial for food safety and public health.

Purpose of the Study:

To develop a sensitive electrochemical aptasensor for Ochratoxin A detection.
To utilize a DNA walker mechanism for signal amplification in OTA sensing.

Main Methods:

Construction of a biosensor using gold nanoparticles-molybdenum oxide nanocomposites.
Integration of hybridization chain reaction (HCR) and a restriction endonuclease-aided DNA walker machine.
Electrochemical detection using differential pulse voltammetry (DPV).

The aptasensor demonstrated high sensitivity for OTA detection.
Achieved a wide linear range (0.01-10000 pg mL⁻¹) and a low detection limit (3.3 fg mL⁻¹).
Showed excellent agreement with UPLC analysis in real samples like juices, wine, and serum.

Conclusions:

The developed aptasensor offers a highly sensitive and reliable method for OTA detection.
The DNA walker amplification strategy significantly enhances biosensor performance.
This method is suitable for practical application in food and biological sample analysis.
