Background:

Intestinal epithelial cells face oxidative stress from various toxins.
Glutathione (GSH) is a key intracellular antioxidant.
Mechanisms of GSH uptake and its protective role in the small intestine are not fully understood.

Purpose of the Study:

To investigate the protective role of exogenous GSH in rat small-intestinal epithelial cells.
To elucidate the mechanism of GSH uptake and its dependence on cellular systems.

Main Methods:

Rat small-intestinal epithelial cells were exposed to t-butyl hydroperoxide or menadione.
Cellular protection was assessed in the presence and absence of exogenous GSH.
GSH uptake was studied using radiolabeled GSH and investigating its dependence on Na+ and membrane potential.

Main Results:

Exogenous GSH significantly protected cells against t-butyl hydroperoxide and menadione-induced injury.
Protection was contingent on the uptake of intact GSH into the cells.
GSH uptake was mediated by a Na+-dependent, electrogenic system located in the basolateral membrane.

Conclusions:

Rat small-intestinal epithelial cells can utilize extracellular GSH to enhance intracellular detoxification.
A specific Na+-dependent transporter facilitates GSH uptake across the basolateral membrane.
This uptake mechanism supports cellular defense against chemically induced oxidative stress.
