Summary

Murine erythroleukemia cells with more DNA attachment sites show higher levels of labeled thymidine in matrix-bound DNA. This suggests that DNA attachment sites are rich in Adenine-Thymine (A+T) base pairs.

Purpose of the Study:

To investigate the relationship between DNA attachment sites and their base pair composition.
To determine if specific DNA sequences are preferentially associated with the nuclear matrix.

Murine erythroleukemia cells were cultured with 3H-thymidine.
DNA was extracted and fractionated to isolate matrix-bound DNA.
Radioactivity (labeled thymidine incorporation) was quantified in both matrix-bound and total DNA.

Cells with a higher number of DNA attachment sites exhibited a greater proportion of labeled thymidine in their matrix-bound DNA compared to their total DNA.
