Summary

Alzheimer's disease pathogenesis may stem from distorted brain capillaries. This leads to reduced nutrient delivery and waste removal, causing neuronal energy crises and contributing to hallmark AD pathology.

Background:

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by progressive cognitive decline.
Current understanding of AD pathogenesis involves amyloid plaques and neurofibrillary tangles.

Purpose of the Study:

To present a working hypothesis on Alzheimer's disease pathogenesis.
To link brain capillary distortion to the development of AD pathology.

Review of ultrastructural and histologic findings in Alzheimer brains.
Hypothesizing the impact of capillary distortion on cerebral blood flow and neuronal function.

Brain capillary distortion alters cerebral blood flow patterns, leading to microturbulence.
